FRIDAY, May 18, 2018 (HealthDay News) — For patients undergoing resection of colorectal liver metastases (CRLM), the presence of the V600E BRAF<\/i> mutation is associated with worse prognosis, according to a study published online May 16 in JAMA Surgery<\/i>.<\/p>\n
Georgios Antonios Margonis, M.D., Ph.D., from the Johns Hopkins University School of Medicine in Baltimore, and colleagues conducted a cohort study to examine the prognostic association of BRAF<\/i> mutations with survival and recurrence independently and compared with other prognostic determinants. Patients who underwent resection for CRLM with curative intent and had data on BRAF<\/i> and KRAS<\/i> mutational status were identified; 849 patients were included in the analyses.<\/p>\n
The researchers found that 5.1 percent of patients had a mutated (mut) BRAF<\/i>\/wild-type (wt) KRAS<\/i> (V600E and non-V600E) genotype, 56.5 percent had a wtBRAF<\/i>\/wtKRAS<\/i> genotype, and 38.4 percent had a wtBRAF<\/i>\/mutKRAS<\/i> genotype. On multivariable analysis, V600E, but not non-V600E, BRAF<\/i> mutation was correlated with worse overall survival and disease-free survival (hazard ratios, 2.76 and 2.04, respectively). Compared with the KRAS<\/i> mutations, the V600E BRAF<\/i> mutation was more strongly associated with overall survival and disease-free survival (\u03b2 for overall survival, 10.15 versus 2.94; \u03b2 for disease-free survival, 7.14 versus 2.27).<\/p>\n
“The presence of the V600E BRAF<\/i> mutation was associated with worse prognosis and increased risk of recurrence,” the authors write.<\/p>\n
